Résumé
In this study, a reporter allowing us to measure the frequency of frameshift mutations in plants was developed. The reporter construct was introduced into the genome of 'Arabidopsis thaliana' via ' Agrobacterium'-mediated transformation. Five single-copy lines were obtained and three of these showed a low, but constant mutation frequency over the T3, T4 and T5 generations. A UV treatment applied to two reporter lines resulted in a significant increase (~18 fold) in the mutation rate in both lines. This response to a DNA damaging agent validated the potential of these lines as tools to measure the frequency of frameshift mutations in plants. The insertion sites of the T-DNA in the reporter lines were finally determined since the lines will eventually be used by crossing them with mutants deficient in different DNA repair mechanisms.
